What L-Theanine Is
L-theanine is an amino acid found in tea. Researchers have studied isolated L-theanine and combinations with caffeine for attention, stress, and sleep, but products and study designs vary.
What One Trial Found
A randomized, placebo-controlled crossover trial in 30 healthy adults studied 200 mg per day for four weeks. Some stress-related, sleep-subscale, and cognitive measures changed, while several overall comparisons were not statistically significant versus placebo. A study this small cannot establish treatment efficacy, long-term safety, or the best dose for the general population. See Source 1 below.
Claims that L-theanine reliably eliminates caffeine side effects, treats anxiety, or improves a sleep disorder are stronger than this evidence supports. Tea also contains varying amounts of caffeine, so drinking tea is not equivalent to taking an isolated supplement.
Safety and Interactions
Dietary supplements are not approved by the FDA for effectiveness before marketing. L-theanine may be combined with caffeine or other active ingredients, and supplement labels and purity can vary. Discuss use with a clinician or pharmacist if you take medicines, are sensitive to caffeine, are pregnant or breastfeeding, or are trying to manage a persistent sleep or mental-health symptom. How much green tea would you need to match a supplement dose?
The answer varies too much by tea and brewing method to give one reliable cup count.
In a 2025 laboratory analysis, brewed green tea averaged 12.5 ± 2.5 mg of L-theanine per 200 mL, but tea type, product, temperature and steeping time all changed the result. Human supplement studies commonly test isolated doses around 100–200 mg, while sleep trials use varied doses and formulations. At the study average, 200 mg would require about 16 cups—not eight to ten—but real tea cannot be dosed precisely from a generic average.
Matcha and shaded teas can contain more L-theanine per serving, but cultivar, powder amount and preparation still create wide variation. "One matcha equals a supplement" is not a dependable conversion unless the product has been analytically tested.
There is no fixed natural theanine-to-caffeine ratio and no single 2:1 research standard. Trials have tested combinations such as 97 mg theanine with 40 mg caffeine and 200 mg theanine with 160 mg caffeine. Those small acute studies cannot justify a universal ratio or a recommendation to supplement every coffee.
Does L-theanine work immediately or does it build up?
If an acute effect occurs, the timing is likely measured in hours rather than weeks—but the evidence does not establish that everyone will feel one.
In a 12-person human pharmacokinetic study, plasma L-theanine peaked about 0.8 hours after a 100 mg dose. That measures absorption into blood, not the time it crosses the human blood–brain barrier or guarantees a subjective effect. Small EEG studies have reported acute changes after single doses, but results and outcome measures vary.
Claims that L-theanine never produces tolerance or that the first dose predicts every later response have not been established in long-term trials. Most cognition studies are acute, which means they cannot answer a chronic-tolerance question.
The practical consequence is more limited: an acute crossover trial can detect short-term group differences, but an unblinded personal comparison is vulnerable to expectation, sleep, caffeine withdrawal and day-to-day variation. Do not treat a single good or bad day as proof.
Set expectations at the right level. Some small trials report improvements on selected attention tasks with caffeine plus L-theanine; others find no advantage for the combination on their primary tasks. It is not an established cognitive enhancer.
Can you take L-theanine with medication?
Short studies are broadly reassuring, but interaction and long-term safety data are limited. There are several uncertainties worth raising with a pharmacist.
Blood pressure medication. Small acute studies sometimes measure blood-pressure changes, but direct drug-interaction evidence is sparse. Anyone taking antihypertensive medication or prone to low blood pressure should ask a pharmacist rather than assuming the combination is harmless.
Sedatives and sleep medication. Claims that L-theanine "promotes GABA" do not establish a clinical interaction with benzodiazepines or sleep medicines. Because combination safety has not been well studied, do not add it to prescription sedatives or alcohol without professional advice.
Stimulant medication for ADHD. People frequently combine L-theanine with prescribed stimulants on the reasoning that it smooths caffeine. The evidence for the caffeine pairing does not automatically transfer to methylphenidate or amphetamine, which act differently, and this is a conversation for a prescriber rather than an experiment.
More generally, small trials have not identified major safety signals at the studied doses, but sample sizes and follow-up are inadequate for a blanket long-term-safety claim. FDA does not approve dietary supplements for safety or effectiveness before sale. A GRAS notice concerns use as a food ingredient under specified conditions; it is not FDA approval of an L-theanine supplement as a treatment. Independent certification can add a quality check but does not prove efficacy.
Pregnancy and breastfeeding have not been studied here, so there is no evidence base in either direction.
